Oyster Mushroom Grain Spawn: Boiled Wheat with Lime and Gypsum, 121 °C

The "mother seed" of oyster mushroom: wheat boiled, limed to about pH 9, sterilised in litre bottles and overgrown from a piece of agar culture. It is the spawn that starts a straw crop. The figures are those of a 2018 study at the Benemérita Universidad Autónoma de Puebla, Mexico, with their strain CP-50 of Pleurotus ostreatus. Source: Valencia de Ita et al., J. Pure Appl. Microbiol. 12(2) 679-686 (2018), p. 680. The bottle closure, the scalpel and the hood are ours. The picture shows an oyster mushroom culture on agar, the starter.

Instructions

1

Boil the wheat

Boil 10 kg of wheat in 15 L of water for 20 minutes. Let it stand 10 minutes, drain it and let it cool at room temperature for 30 minutes.

Bottle and sterilise

Pack the grain into 20 bottles of 1 L, close them, and sterilise them for 60 minutes at 121 °C. Leave them 24 hours to cool completely.

Inoculate from agar

Working in a clean-air hood, cut a piece of agar colonised by the oyster mushroom with a flamed scalpel and drop it into each bottle. The study used 22.5 cm² per bottle.
